RNase A is an endoribonuclease that specifically hydrolyzes RNA 3´ of pyrimidine residues and cleaves the phosphodiester linkage to the adjacent nucleotide. RNase A is a single chain polypeptide containing 4 disulfide bridges. In contrast to RNase B, it is not a glycoprotein. RNase A is used to remove RNA during procedures for the isolation of plasmid and genomic DNA. It is a highly purified preparation of bovine pancreatic ribonuclease A suitable for use in selective removal of RNA.
RNase is inhibited in the presence of heavy metal ions. RNase is also inhibited competitively by DNA. Its optimum temperature and pH are 60℃ and 7.6 respectively, and it is active between pH=6-10. The final concentration of RNase A in the plasmid is 100μg/ml, 10 mg/ml RNaseA is added to the plasmid solution according to 1/100 volume.
